Detection of a single nucleotide polymorphism (SNP) DNA marker linked to cocoon traits in the mulberry silkworm, Bombyx mori (Lepidoptera: Bombycidae)

Cocoon weight and shell weight are the key economic traits ultimately determining silk yield. In order to detect the main quantitative trait loci (QTL) associated with the cocoon traits of the mulberry silkworm, Bombyx mori, the parents of larvae that produced cocoons that differed greatly in weight and shell weight were screened using 240 primer pairs of single nucleotide polymorphic markers (SNPs) representing all the 28 linkage groups in silkworm. Out of the 240 primer pairs, 48 (20%) revealed distinct polymorphism between the parents, which was confirmed by the co-dominant expression of both polymorphic PCR products in the F1 generation. The bulked segregant analysis (BSA) was used to compare the SNP profiles of the parents, F1 and F2 bulks using the 48 informative SNP primers. This revealed that out of 48 primer pairs, only one pair, i.e., No. 04124 of the linkage group 4 showed clear differences in the amplified products between the bulks corresponding to that of the parents with different cocoon traits suggesting that the DNA regions amplified by this primer pair are closely linked to the QTL controlling the cocoon traits. The results were also confirmed by screening the backcross (BC) progeny. This is the first report of the identification of a QTL using SNPs with BSA. The results of the present study indicate that it might be possible to use SNPs for marker assisted selection (MAS) in silkworm breeding programs aimed at improving cocoon traits.

Phylogeny and nomenclature of the box tree moth, Cydalima perspectalis (Walker, 1859) comb. n., which was recently introduced into Europe (Lepidoptera: Pyraloidea: Crambidae: Spilomelinae)

The box tree moth, Cydalima perspectalis (Walker, 1859) comb. n., is native to India, China, Korea, Japan and the Russian Far East. Its larvae are a serious pest of different species of Buxus. Recently, C. perspectalis was introduced into Europe and first recorded from Germany in 2006. This species has been placed in various spilomeline genera including Palpita Hübner, 1808, Diaphania Hübner, 1818, Glyphodes Guenée, 1854 and the monotypic Neoglyphodes Streltzov, 2008. In order to solve this nomenclatural confusion and to find a reasonable and verifiable generic placement for the box tree moth, the morphology of the above mentioned and some additional spilomeline taxa was investigated and their phylogeny analysed. The results show that C. perspectalis belongs to a monophylum that includes three of the genera in which it was previously placed: Glyphodes, Diaphania and Palpita. Within this monophylum, it is closely related to the Asian Cydalima Lederer, 1863. As a result of this analysis, Sisyrophora Lederer, 1863 syn. rev. and Neoglyphodes Streltzov, 2008 syn. n. are synonymised with Cydalima Lederer, 1863, and five species are transferred to this genus: Cydalima capriniodes (Hampson, 1912) (Glyphodes) comb. n., Cydalima decipiens (Hampson, 1912) (Glyphodes) comb. n., Cydalima joiceyi (Janse, 1924) (Margaronia) comb. n., Cydalima perspectalis (Walker, 1859) (Phakellura) comb. n. and Cydalima pfeifferae (Lederer, 1863) (Sisyrophora) comb. rev.

Changes in the life history traits of the European Map butterfly, Araschnia levana (Lepidoptera: Nymphalidae) with increase in altitude

Climatic conditions can modify the life history traits, population dynamics and biotic interactions of species. Therefore, adaptations to environmental factors such as temperature are crucial for species survival at different altitudes. These adaptive responses, genetically fixed or plastic (phenotypic plasticity), can be determined by physiological thresholds and might vary between sexes. The objective of this study was to determine whether the life history traits of the European Map butterfly (Araschnia levana) differ at different altitudes. A field experiment was carried out along an altitudinal gradient from 350 to 1010 m a.s.l. in a low mountainous region (Bavaria, Germany). 540 butterfly larvae were placed at different altitudes in 18 planted plots of their larval host plant, the stinging nettle (Urtica dioica). After three weeks the larvae were collected and reared under laboratory conditions. Developmental traits of the butterflies, mortality and percentage parasitism were measured. Larval development was generally slower at higher altitudes and lower temperatures and larval weight decreased with increasing altitude and decreasing temperature. However, there were no significant differences in pupation, adult lifespan and percentage mortality at the different altitudes and temperatures. Female larvae were heavier than those of males, and the pupal and adult lifespans were longer in females than in males. However, male and female butterflies reacted similarly to altitude and temperature (no significant interactions). None of the 188 larvae collected were parasitized. In conclusion, the phenotypic plasticity of European Map butterfly has enabled it to adapt to different temperatures, but the strategies of the sexes did not differ.

Biology of the conifer needle scale, Nuculaspis abietis (Hemiptera: Diaspididae), in northern Iran and parasitism by Aspidiotiphagus citrinus (Hymenoptera: Aphelinidae)

The conifer needle scale, Nuculaspis abietis (Schrank) emerged as an important pest of conifers in the Kelardasht region of Mazandaran province, Iran, in the late 1990's. This pest feeds on conifer needles and twigs causing needle drop and branch desiccation. Its discovery in Kelardasht in Mazandaran Province necessitated a local quarantine of conifers in the genera Picea, Abies, and Pinus. We studied the life history and ecology of this scale on Norway spruce, Picea abies, under laboratory and natural field conditions. The complete life cycle of females required 206.4 ± 5.7 days in the laboratory (25 ± 1°C, 65-75% RH, 14L : 10D) and 315.7 ± 9.3 days under field conditions. In Kelardasht, numbers of adult males and females peaked in mid-June and early May, respectively, and numbers of first and second instar nymphs in mid-July and early September. Lifetime fecundity was estimated to be 57.3 ± 5.1 eggs and 54.0 ± 4.4 nymphs per female under laboratory conditions. The sex ratio ranged from 59% female for second instar nymphs to 71% female for adults. Aspidiotiphagus citrinus (Crawford) (Hymenoptera: Aphelinidae) was found naturally parasitizing the scale and overwintering in the larval stage on second instar nymphs. First generation adult wasps emerged in spring from overwintered second instar nymphs to parasitize 64.75% of first instar scales. Second generation wasps emerged from early September to mid-October and parasitized 19.75% of second instar scales, for a cumulative parasitism rate of 84.5%.

Relationship between the development of Simulium maculatum (Diptera: Simuliidae) and the thermal regime

Warmer temperatures in the past 30 years have significantly influenced the seasonal development of insects throughout Europe. As a result of the outbreaks of black flies that have occurred in southeastern Lithuania since the 1970s it is hypothesized that this increase in black fly activity is due to the change in climate. To test this hypothesis the development of Simulium maculatum Meigen under different conditions was determined. This revealed that the time of hatching of S. maculatum eggs in Lithuania was influenced by winter air temperatures, especially those in March. Pupation in S. maculatum is associated with the increase in air and water temperatures that occur at the end of April and in May. The emergence of S. maculatum black flies occurs most often in May. At a water temperature of 13.2°C (1999), S. maculatum took 42 days to develop and half this time (21 days) when the water temperature was 18.8°C (2005). The number of black flies that emerged each year was determined by air temperature but unaffected by rainfall in June (either per month or per ten-day period).

Recombinant expression, purification and characterization of Bombyx mori (Lepidoptera: Bombycidae) pyridoxal kinase

Pyridoxal kinase (PLK; EC 2.7.1.35) is a key enzyme in the metabolism of vitamin B6 (VB6) in Bombyx mori. A fusion expressional vector pET-22b-BPLK-His was constructed using a sub-cloning technique, the recombinant B. mori PLK was then expressed in Escherichia coli, purified and characterized. Bioinformatics were used to deduce the protein structure and genomic organization of this enzyme. Using Ni Sepharose affinity column chromatography, the recombinant protein was purified to very high degree (approximately 90%). The recombinant PLK exhibits a high specific enzymatic activity (1800 nmol/min/mg of protein). The maximum catalytic activity of this enzyme was recorded over a narrow pH range (5.5-6.0) and Zn2+ is the most effective cation for catalysis under saturating substrate concentrations. When only triethanolamine is present as the cation, K+ is an activator of PLK. A double reciprocal plot of initial velocity suggests that the enzyme catalyses the reaction by means of a sequential catalytic mechanism. Under optimal conditions, the Km value for the substrates of ATP and pyridoxal are 57.9 ± 5.1 and 44.1 ± 3.9 µM. B. mori's genome contains a single copy of the PLK gene, which is 7.73 kb long and contains five exons and four introns, and is located on the eighth chromosome. The PLK may be a dimer with two identical subunits under native conditions, and it is hypothesized that each monomer contains eight α-helices (α1-8), nine β-strands (β1-9) and two segments of 310 helices.

Determining the season of death from the family composition of insects infesting carrion

Determining the season of death by means of the composition of the families of insects infesting carrion is rarely attempted in forensic studies and has never been statistically modelled. For this reason, a baseline-category logit model is proposed for predicting the season of death as a function of whether the area where the carcass was exposed is sunlit or shaded and of the relative abundance of particular families of carrion insects (Calliphoridae, Fanniidae, Sarcophagidae, and Formicidae). The field study was conducted using rodent carcasses (20-252 g) in an urban forest in southeastern Brazil. Four carcasses (2 in a sunlit and 2 in a shaded area) were placed simultaneously at the study site, twice during each season from August 2003 through June 2004. The feasibility of the model, measured in terms of overall accuracy, is 64 ± 14%. It is likely the proposed model will assist forensic teams in predicting the season of death in tropical ecosystems, without the need of identifying the species of specimens or the remains of carrion insects.

Analysis of the lipids mobilized by adipokinetic hormones in the firebug Pyrrhocoris apterus (Heteroptera: Pyrrhocoridae)

Effects of two adipokinetic hormones (Pyrap-AKH and Peram-CAH-II) on the presence of diacylglycerol (DG) molecular species and their fatty acid (FA) constituents in the haemolymph of the firebug Pyrrhocoris apterus were investigated using liquid chromatography (HPLC) and electrospray ionization mass spectrometry (ESI-MS). The results show that DGs with characteristic FAs are preferentially mobilized from the fat body (FB) by the action of both the AKHs produced by P. apterus. Both the macropterous and brachypterous morphs have similar DG and FA profiles. A difference in the action of the Pyrap-AKH and the Peram-CAH-II, however, results in distinct differences in the distribution of FAs in the macropterous morph. It seems that C16 to a slight extent and unsaturated C18 FAs mainly play a dominant role in the AKH based action, in particular linoleic acid (18:2), which represents 50-60% of the total DG mobilized. The metabolically active C16 and C18 FAs are preferentially absorbed from the linden seeds and accumulated in the FB. The relationships between AKH action and FA distribution in DGs in P. apterus, compared to other insect species are summarized and discussed in detail.

Fluorimetric determination of hydrogen peroxide production by the haemocytes of the wax moth Galleria mellonella (Lepidoptera: Pyralidae)

The aim of this study was to investigate whether haemocytes of Galleria mellonella (Lepidoptera: Pyralidae) larvae produce reactive oxygen species (ROS) like human blood phagocytes. The production of ROS was measured first using luminol-enhanced chemiluminescence of un-stimulated and stimulated (four activators with different modes of action) haemolymph or isolated haemocytes. However, spontaneous and activated production of ROS remained at the background level. In subsequent experiments an ultrasensitive fluorescence method using Amplex Red reagent to detect hydrogen peroxide (H2O2) was used. After optimization, Amplex Red was successfully used for determining H2O2 production by both un-stimulated and stimulated haemocytes. To determine the affect of pH and ions on the measurement, several diluent solutions were tested. This revealed that Ca2+ and Mg2+ ions are less important for the reaction in insect than mammalian cells. Among the activators tested, phorbol myristate acetate (PMA) and calcium ionophore (Ca-I) had the best stimulatory effect on insect samples, while opsonised zymosan particles (OZP) was the best activator for human phagocytes. In conclusion, the haemocytes of G. mellonella produce H2O2 as an important innate immunity factor, but under different conditions and in different amounts, which probably results in them being less effective in killing microbes than human phagocytes.

Genetic structure and phenotypic diversity of two northern populations of Cheilosia aff. longula (Diptera: Syrphidae) has implications for evolution and conservation

The genetic structure and phenotypic diversity of two populations of Cheilosia aff. longula (Diptera: Syrphidae) in Lapland, Finland, were examined using DNA sequencing, protein electrophoresis, and geometric morphometrics. The morphological identification of the species were verified using partial sequences of mitochondrial cytochrome c oxidase subunit I (COI mtDNA), and the nuclear ribosomal internal transcribed region 2 (ITS2 rDNA), and comparing the corresponding sequences of Cheilosia aff. longula and the closely related C. longula. Two and three haplotypes of the genes COI mtDNA and ITS2 rDNA were identified in the two populations. Analysis of 12 isozyme loci showed an extremely low genetic variability in the populations originating from Utsjoki and Kevo. Discriminant analysis combined with canonical variate analysis revealed inter-population divergence in wing shape. Variation among genetically diverse individuals, both within- and among studied populations was studied, and directional (DA) and fluctuating asymmetry (FA) estimated using landmarks in the framework of geometric morphometrics. It is likely that the documented DA and FA asymmetry in both wing shape and size reflects the developmental instability of the individuals studied. By using Procrustes ANOVA the locations of particular landmarks responsible for the variation in shape were determined. The decomposition of the components of variance accorded to each landmark showed that the landmarks differed in the percentage of variation they accounted for (DA, FA and variation among individuals). In the discussion the implications of the reduced genetic diversity and asymmetry in wing traits for conservation is considered.

Estimations of the critical temperatures for development of the pistachio psylla, Agonoscena pistaciae (Hemiptera: Psyllidae)

The pistachio psylla, Agonoscena pistaciae Burckhardt & Lauterer (Hemiptera: Psyllidae), is a major pest of pistachio trees throughout the pistachio producing regions in Iran. The effect of temperature on the developmental rates of eggs and nymphs of A. pistaciae was determined at different constant temperatures, i.e. 15, 20, 25, 30, 32.5 and 35 ± 0.5°C. The relationships between temperature and developmental rates were described by linear and the non-linear Lactin models. These models were evaluated based on R2, RSS, AIC and R2adj. The estimated value of the lower temperature threshold for egg, nymph and egg to adult development based on the linear model was 8.06, 10.38 and 9.97°C, respectively, and based on the Lactin model was 8, 11.55 and 11.2°C, respectively. Thermal constants estimated using the linear model, were 88.5, 243.90 and 333.33 DD, respectively, for egg, nymph and egg to adult development. These results indicate that the linear model gives a better description of the relationship between developmental rate and temperature for A. pistaciae than the non-linear model. These results could be incorporated into forecasting models used in the integrated pest management of this pest.

Alternative foods for the multicoloured Asian lady beetle Harmonia axyridis (Coleoptera: Coccinellidae)

The multicoloured Asian lady beetle Harmonia axyridis (Coleoptera: Coccinellidae) may potentially exploit a range of "alternative" food sources. The nutritional value of three common non-prey food groups, i.e. fruit, fungi and pollen, is examined in this study. Development, reproduction and survival of the species were assessed in the laboratory on diets of apple, pear and raspberries and the fungi Oidium lycopersicum, Botrytis cinerea, Sclerotinia sclerotiorum and Rhizoctonia solani. When fed exclusively on these foods this ladybird failed to complete its development or reproduce. However, larval and adult survival was prolonged by 4 to 8 days and 55 to 67 days, respectively, when fruit was offered compared with only water. During a field study H. axyridis adults were collected monthly from March to October at two locations near Ghent, Belgium. Gut analysis demonstrated that, despite the continued presence of aphids, over 90% of all the adults collected contained pollen throughout the year. The pollen belonged to 53 different pollen types. Monthly average numbers of pollen grains in the gut varied from 5 to 233 grains. In contrast, there were about 35,000 grains in the guts of H. axyridis females that successfully developed and reproduced in the laboratory on diet that consisted of only pollen. This suggests that in the field in the majority of cases pollinivory mainly only provided complementary nutrients for this coccinellid. The use of alternative non-prey foods like pollen and fruit may play a role in sustaining populations of this coccinellid at times when the optimal prey is absent.

Cohort-splitting in the millipede Polydesmus angustus (Diplopoda: Polydesmidae): No evidence for maternal effects on life-cycle duration

Under seasonal conditions, Polydesmus angustus individuals born in the first part of the breeding season have a 1-year life cycle and those born later have a 2-year life cycle (cohort-splitting). In this study, 249 juveniles from four early broods (born in mid-July) and four late broods (born in September) were reared under similar laboratory conditions, to test for possible maternal influences on life-cycle duration. Development times of early- and late-born individuals were compared under four combinations of day length and temperature (16 h - 18°C, 16 h - 16°C, 12 h - 18°C and 12 h - 16°C). The results showed that development time varied significantly in response to day length, temperature and sex, but that of individuals in the early and late broods did not differ significantly (mean development times ± SE: 180 ± 6 and 183 ± 8 days, respectively). There were no significant interactions between birth period and other factors, indicating that the effects of day length, temperature and sex on development time were similar in early- and late-born individuals. This indicates that the extended life cycle of millipedes born late in the season is not maternally determined and that cohort-splitting is controlled entirely by the environmental conditions experienced by the offspring during their development. This conclusion is supported by the absence of significant variation in offspring live weight at birth measured at different times in the breeding season. The results are discussed in relation to the bet-hedging theory, which is often put forward to account for cohort-splitting in arthropods. In P. angustus, the results are consistent with either bet-hedging or adaptive plasticity, but further studies are required to decide which interpretation is correct.

Parasitylenchus sp. (Tylenchomorpha: Allantonematidae) parasitizing field populations of Harmonia axyridis (Coleoptera: Coccinellidae)

Adults of the invasive harlequin ladybird, Harmonia axyridis (Coleoptera: Coccinellidae), were found to be parasitized by nematodes (Tylenchomorpha: Allantonematidae) in Denmark. The nematodes were identified as Parasitylenchus sp. Major morphological characters of the nematodes did not differ significantly from Parasitylenchus coccinellinae Iperti & van Waerebeke 1968, but minor deviations in egg dimension and tail length were measured. Parasitism was only recorded in early autumn, with prevalence ranging from 2 to 33%. Adult and juvenile nematodes occurred together in the body cavity of both female and male H. axyridis.

The dispersal ability of wood cricket (Nemobius sylvestris) (Orthoptera: Gryllidae) in a wooded landscape

Information on the dispersal ability of flightless insect species associated with woodlands is severely lacking. Therefore, a study was conducted examining the dispersal ability of wood cricket (Nemobius sylvestris) juveniles (nymphs) and adults in a wooded landscape on the Isle of Wight, UK, to further our understanding of the ecology and management of this and other flightless insects. A series of experiments were conducted where nymphs and adults were released and observed at a range of spatial-temporal scales within different habitat environments. The results of this investigation indicated no difference in the dispersal ability of wood cricket nymphs and adults. However, adult females moved less than adult males. Adult males were found to spread twice as fast as females, and males were found capable of traversing up to 55 m away from woodland habitat through semi-natural and grazed grassland. Additionally, rates of population spread of both wood cricket nymphs and adults (males and females) were found to be accurately described by the inverse-power function, predicting only few individuals dispersing over time. Together this indicates that overall colonization success and rate of population spread, being female dependent, is likely to be low for this species. Wood crickets were found to live in and able to move along mature woodland edges directly bordering agricultural land. This indicates that conservation initiatives focusing on creating woodland corridors and developing woodland habitat networks have the potential to facilitate the spread and population viability of wood crickets, if the woodland edges are given enough time to develop. However, long-term monitoring is needed to evaluate the overall effectiveness of such approaches.

Influence of male density on guarding behaviour in the water strider Aquarius paludum (Heteroptera: Gerridae)

In species with last sperm precedence, post-mating guarding behaviour by males is effective for avoiding sperm displacement by additional males. However, for males to guard their mates under all circumstances may be undesirable, particularly when the population density of conspecific rival males is low. To better understand the effects of rival density on mate guarding behaviour, we compared post-copulatory behaviour in males artificially subjected to different male densities using Aquarius paludum. Our results indicate that males of A. paludum modulate their post-copulatory behaviour based on previously experienced rival densities.

Some ethological aspects of the trophobiotic interrelations between ants (Hymenoptera: Formicidae) and larvae of the sawfly Blasticotoma filiceti (Hymenoptera: Blasticotomidae)

Some ethological aspects of the interrelations between ants and the larvae of Blasticotoma filiceti Klug, 1834 were investigated in the Altai Republic and Novosibirsk Region in 2006-2008. The interactions of ants with the larvae of this sawfly are determined by the concealed way of life style of B. filiceti. The majority of the ant-sawfly encounters occurred near holes in fern fronds at the moment when larvae excreted liquid or left their tunnels before descending to the soil prior to overwintering. Sawfly larvae visited by more aggressive ants, such as Formica s. str., leave the fern fronds slowly, which enables them to avoid inciting attacks by ants. The behaviour of the ants while collecting the larval excretion is similar to their behaviour at sugar troughs. The organisation of the collecting larval excreta by ants was investigated in detail in the cases of Formica polyctena Förster, 1850 and Myrmica rubra Linnaeus, 1758. The individual fern plants with sawfly larvae are attended by relatively constant groups of foragers in both cases. However, the highly social red wood ants interact with sawfly larvae in a more complex way. While the working groups of M. rubra tending sawfly larvae consist only of non-aggressive "unspecialized" foragers, those of F. polyctena include also a few "on duty" ants that protect the trophobionts, at least from the other ants.

Does climate influence phenological trends in social wasps (Hymenoptera: Vespinae) in Poland?

Responses of insects to recent climate change have been well documented in a number of taxa, but not in wasps. This study examined shifts in phenology of the two most important wasp species (Vespa crabro and Vespula germanica) in Poland over the last three decades. Both species showed similar temporal trends, advancing their phenology after the early 1980s, but this pattern was detected only for workers not for the appearance of queens. The appearance times for V. germanica were negatively related to mean April temperature, appearing earlier in years with warmer springs, and positively related to precipitation in April. The studied species advanced aspects of their phenology, but linking this to temperature was not achieved for V. crabro suggesting that we have to pay more attention to the life history traits of the study organisms.

Feeding conditions modify the photoperiodically induced dispersal of the water strider, Aquarius paludum (Heteroptera: Gerridae)

Overwintering adults of the water strider, Aquarius paludum were collected from the field in fall and kept under short days (12L : 12D) at 20°C for a week. A control group was then kept at 12L : 12D and fed daily, and three experimental groups were transferred to 15.5L : 8.5D and fed at different frequencies: daily or every 2nd or 3rd day. Temperature was kept at 20°C. Flight behaviour of the adults in the four groups was recorded every 10 days up to the end of the 5th week when the adults were dissected. State of the flight muscles and reproductive organs (ovaries and testes) was recorded. Flight ability of the adults fed every 3rd day (Group 3) was higher than that of those fed daily (Group 1). Seventy percent of the females that were fed every 3rd day (Group 3) remained in diapause for 5 weeks, in spite of the reproduction promoting long-days, while none of the females fed every day remained in diapause (Group 1). Flight muscle histolysis, promoted by long-days, was inhibited by poor feeding conditions. Thus, they can migrate and seek more suitable water bodies rich in food. The modifying effect of the availability of food on the trade-off between reproduction and flight is recorded here for the first time for a carnivorous insect.

Molecular differentiation of the four most commonly occurring Trichogramma (Hymenoptera: Trichogrammatidae) species in China

Trichogramma dendrolimi
, T. ostriniae, T. confusum and T. evanescens are the four most commonly occurring Trichogramma species with overlapping distribution in China. They are the most frequently used egg parasitoids for biological control of lepidopterous crop pests in China. It is difficult to differentiate Trichogramma species because of their small size and lack of differences in morphological characters. Different molecular markers were employed to molecularly characterize and differentiate these species, including direct amplification of the internally transcribed spacer 2 (ITS2) of ribosomal DNA by polymerase chain reaction (PCR), restriction fragment length polymorphism (RFLP), random amplified polymorphic DNA (RAPD) and species-specific primers. The results showed that direct amplification of ITS2 could not clearly discriminate these species, but they could be differentiated using RFLP pattern obtained with endonucleases EcoRI and HindIII. The banding pattern produced by RAPD is irreproducible so it is not a suitable way to identify Trichogramma species. Finally, the species-specific primers designed based on ITS2 sequences could unequivocally distinguish the four species. The species-specific primer-based protocol proved to be the most convenient and time saving method for the identification of Trichogramma species by creating a unique PCR product, which can be used in surveying natural populations of Trichogramma species. This is the first report of the prompt identification of the four most commonly occurring Trichogramma species in China.

Effect of temperature on the life history of Encarsia bimaculata (Hymenoptera: Aphelinidae), a parasitoid of Bemisia tabaci (Hemiptera: Aleyrodidae)

Encarsia bimaculata
(Heraty & Polaszek) is an abundant parasitoid of Bemisia tabaci in southern China. The effects of constant temperatures on a range of life history traits, including development, survival of immatures, longevity and reproduction of adults, were studied in the laboratory. The developmental period from egg to adult ranged from 34.3 ± 0.4 d at 20°C to 8.7 ± 0.6 d at 32°C, A total of 181.4 ± 2.4 degree-days were required to complete development with a lower developmental threshold of 11.6 ± 0.3°C. The survivorship of E. bimaculata from 2nd instar to adult varied from 81.3 ± 1.7% at 20°C to 91.0 ± 1.8% at 26°C. Average adult female longevity was 8.4 ± 0.7 d at 20°C and 5.4 ± 0.4 d at 32°C, and daily production of offspring peaked at 29°C with 4.5 offspring per female. The maximum oviposition occurred three days after adult emergence at 23, 26, 29 and 32°C, and four days at 20°C. Total number of offspring produced per female varied from 24.3 ± 2.0 at 32°C to 29.3 ± 2.9 at 20°C. The maximum intrinsic rate of increase (rm) was 0.2163 ± 0.013 at 29°C, followed by 0.2062 ± 0.022 at 32°C. Results indicate that E. bimaculata reaches its maximum biological potential at temperatures ranging from 26°C to 32°C with 29°C being the optimal temperature.

Comparative genetic studies of native and introduced Coccinellidae in North America

During the past four decades, several species of aphidophagous Coccinellidae became established in North America, including Coccinella septempunctata, Harmonia axyridis, Hippodamia variegata, and Propylea quatuordecimpunctata. After their establishment, unknown circumstances favoured a rapid increase in population densities and distribution of H. axyridis and C. septempunctata at localities hundreds and thousands of kilometers from their release sites. Propylea quatuordecimpunctata and Hippodamia variegata have spread more slowly after becoming established in northeastern North America. Comparative studies based upon allozyme variation in these four introduced species and in six native North American species of ladybird beetles revealed no significant differences in genetic diversities. Genetic variation, assessed by allelic diversity and heterozygosity, was uncorrelated with the establishment and spread of these predatory species in North America. All ladybirds studied show a remarkable degree of dispersion with little detectable population subdivision.

Kairomone involvement in the host specificity of the egg parasitoid Trissolcus basalis (Hymenoptera: Scelionidae)

This paper reports the results of a comparative laboratory analysis of the behavioural responses of the egg parasitoid Trissolcus basalis (Wollaston) (Hymenoptera: Scelionidae) to semiochemical cues from four species of pentatomid bugs, Nezara viridula (L.), Eurydema ventrale Klt., Murgantia histrionica Hahn. and Graphosoma semipunctatum F. (Heteroptera: Pentatomidae). In a Y-tube olfactometer, T. basalis was attracted by volatile chemicals from N. viridula, but not from other pentatomid species. In an open arena, the parasitoid reacted to chemical trails left on filter paper by all the species but most intensely to those left by N. viridula. However, upon encountering pentatomid eggs, T. basalis examined more intensely and probed more frequently the eggs of G. semipunctatum than those of the other species. The parasitoid only parasitized and emerged from eggs of G. semipunctatum and N. viridula; those of the other species were unsuitable. Therefore N. viridula is semiochemically confirmed to be a coevolved host (old association) of T. basalis, whereas G. semipunctatum may be a potential non-coevolved host (new association). The utility of these tests for defining a parasitoids' host specificity and in assessing the risk of non-target effects in biological control is discussed.

Effects and interactions of temperature, host deprivation and adult feeding on the longevity of the parasitoid Venturia canescens (Hymenoptera: Ichneumonidae)

A laboratory study was carried out to determine the effects and interactions of temperature, host deprivation and adult feeding on the longevity of the parasitoid Venturia canescens (Gravenhorst) (Hymenoptera: Ichneumonidae). The effect of body size was also examined. Large wasps (hind tibia length > 1.96 mm) lived significantly longer than smaller conspecifics (hind tibia length < 1.89 mm). Adults reared at 15°C lived longer regardless of whether supplied with hosts or food. Correspondingly, adults had a shorter life at 30°C. Honey-fed adults lived significantly longer than starved adults at all temperatures and irrespective of host presence, while access to hosts resulted in a decrease in longevity at all temperatures, regardless of food supply. The Weibull distribution was used to describe the age specific survival, which in V. canescens is of "Type I", as the risk of death increases with age. Despite the significant effect of host presence on survival it was less than either temperature or feeding. The interactions between feeding, host presence and temperature proved to be significant. The objective of the study was to improve the effectiveness of V. canescens as a biological control agent of stored product pests.

Activity of Lygus lineolaris (Heteroptera: Miridae) adults monitored around the periphery and inside a commercial vineyard

The tarnished plant bug, Lygus lineolaris (Heteroptera: Miridae), is a highly polyphagous pest that feeds on a broad range of economically important crops in North America. Flying L. lineolaris adults can move from crop to crop rapidly and easily. Little is known about the movement of L. lineolaris in or near vineyards. From May to October 2002 and 2003, 39 white sticky traps were positioned inside and at the periphery of a vineyard to study the movements of tarnished plant bug adults. Tarnished plant bug captures were most numerous from the end of July to mid August, with captures of ca. 3000 individuals in one week. During the vegetative season, most individuals were captured flying in and around the vineyard at a height of between 40 and 60 cm. The frequency with which the weeds were mown affected the numbers of adults captured. At the periphery of the vineyard, tarnished plant bug was more abundant near perennial vegetation, which tends to be stable (an apple orchard, a spruce wood), than in the vineyard where agronomic activities changed the availability of food. The tarnished plant bug adults did not hibernate in the vineyard. It is suggested that appropriate weed management will reduce the abundance of tarnished plant bugs in vineyards as it would deprive them of a number of suitable hosts for feeding and oviposition.

Genetic polymorphism in "mixed" colonies of wood ants (Hymenoptera: Formicidae) in southern Finland and its possible origin

Wood ant colonies that appear to consist of individuals representing different species are described in several previous papers. The present study is the first to elucidate the genetic basis of the spectacular morphological variability observed within such colonies. Two seemingly mixed colonies (FM-1 and FM-2) from southern Finland were investigated. On the basis of the morphology of their workers these colonies were comprised of individuals with phenotypes typical of Formica rufa L., F. polyctena Först., and F. aquilonia Yarr. The sequence of an mtDNA fragment (5' end of the cytochrome b gene) was used to examine the phylogenetic relationships among haplotypes of workers from homogeneous colonies of different wood ant species and the two supposedly mixed colonies, and to sort the individuals within the colonies into matrilines. Six microsatellite loci were used to analyse the genetic differentiation between colonies and among workers within colonies, and to detect putative hybrids. The results show that, independently of their phenotype, workers from the "mixed" colonies were genetically more similar to other individuals in their colony than to those in the homogeneous F. rufa, F. polyctena or F. aquilonia colonies. However, while colony FM-1 consisted of offspring of the same queen or more likely several maternally related queens, colony FM-2 consisted of the offspring of at least four unrelated queens. The data suggest hybridisation between F. polyctena × F. aquilonia and F. polyctena × F. rufa (and possibly subsequent mating between these hybrids) as the most probable mechanism leading to the existence of these two colonies, which implies that the hybrids are fertile. This study shows that colonies of wood ant hybrids can arise spontaneously and persist under natural conditions. The results also revealed that even some morphologically homogeneous colonies are genetically heterogeneous. In the case of closely related, morphologically similar species that interbreed, morphology can be a bad predictor of genetic differences between individuals.

Physiology of heartbeat reversal in adult Drosophila melanogaster (Diptera: Drosophilidae)

Heartbeat patterns were monitored in the living bodies of decapitated adult flies using several electrocardiographic methods (pulse-light optocardiography, thermocardiography, strain-gauge cardiography). Unlike other insect species, in which there is a peristaltic segmental propagation of cardiac contractions, Drosophila uses extremely efficient synchronic cardiac contractions. The rate of synchronic cardiac pulsation, which is characterized by simultaneous propagation of anterograde systolic contractions along all the segments of the heart, is relatively fast (~ 4 Hz at room temperature). This pulsation is used mainly for the vigorous pumping of haemolymph into the head and thorax through a narrow elastic tube, the aorta (anterograde I heartbeat). In addition, this synchronic pulsation is also used to enhance the circulation of haemolymph throughout the abdominal body cavity (anterograde II heartbeat). The switch between thoracic (anterograde I) and abdominal (anterograde II) haemolymph circulation is regulated by periodically alternating, tetanic contractions and relaxations of the conical heart chamber (ventricle). In the latter there is a pair of slit-like apertures, which are closed or opened by contraction or relaxation of the organ, respectively. During contraction of the conical chamber, the apertures are tightly constricted for several seconds and haemolymph is pumped forwards into the aorta (anterograde I heartbeat). Conversely, during relaxation of the conical chamber, the apertures are wide open for a few seconds, haemolymph leaves the heart and leaks out through open apertures and circulates from the tail to the base of the abdomen. The backward oriented, retrograde heartbeat recorded in other insects, has a lower frequency (1 to 2 Hz), occurs in Drosophila only sporadically and usually in the form of individual or twinned systolic peaks of large amplitude. Unlike the synchronic nature of the anterograde I and II cardiac contractions, propagation of the relatively slow retrograde heartbeat is by peristalsis. The newly discovered, compact ventricle with atrium and synchronic functioning of the insect heart shows structural and functional analogies with the functioning of the human heart.

Mesostigmatid mites associated with the dung beetle Copris lunaris (Coleoptera: Scarabaeidae)

We examined the mesostigmatid mites found in four nest chambers of the dung beetle Copris lunaris (Scarabaeidae) in Slovakia. A total of 763 mites was found, belonging to ten species. The most frequent and abundant species were Pelethiphis opacus, Macrocheles copridis, Parasitus copridis, Uropoda copridis, Copriphis pterophilus, and Onchodellus hispani. The nests contained 19 dung balls, each enclosing a beetle pupa. Altogether 472 mites were found in these brood balls. A further 291 mites were found on the parental beetles in the nests. Three mite species were clearly more abundant in brood balls than on parental beetles, and these belonged to the life cycle stage that disperses by phoresy. The mites found in brood balls apparently disperse on the young adult beetles when they emerge. Only Parasitus copridis was more abundant on parental beetles than in brood balls. Different species of mites have developed different strategies for dispersal, as shown by their preferential attachment to either the parental or progeny generations of beetles.

An improved method for the culture of wing discs of the wingless bagworm moth, Eumeta variegata (Lepidoptera: Psychidae)

Adults of the wingless bagworm moth, Eumeta variegata, show remarkable sexual dimorphism. Final-instar larvae of the male have invaginated wing discs, whereas those of the female are rudimentary. To determine the best method for culturing the wing discs of E. variegata, which in both sexes are attached to the larval integument, two methods of culturing the larval wing discs are compared. Initially, a stationary culture was used. In these cultures necrotic cells and degeneration of wing discs of males were sporadically observed. By contrast, many small vacuoles were observed in the female wing rudiment under these conditions. In order to overcome some of the problems associated with stationary culture, rotating culture was used and resulted in the wing discs of males and females remaining in good condition. A histological analysis revealed that the wing disc morphology was normal when they were cultured in this way. These results indicate that rotating the culture medium is the better procedure for studying the action of hormones on the differentiation and metamorphosis of reduced wing rudiments in E. variegata.

Present conservation status of red wood ants in north-western Belgium: Worse than previously, but not a lost cause

The number of colonies of red wood ants (Formica rufa group) in the north western part of Belgium has declined by more than half over the past 20 years. In the study area of 20 × 50 km there are currently only 36 colonies of two species of red wood ants (Formica rufa and F. polyctena). Both species form both monodomous and polydomous colonies. Monodomous F. rufa colonies occur in large areas of open woodland dominated by Pinus sylvestris. The other three types of red wood ant (F. rufa polydomous, F. polyctena mono- and polydomous) occur in clearings or along the margins of small areas of deciduous or mixed woodland. The extinction of red wood ants cannot be attributed to a loss of their main habitat. Their decline in abundance, however, appears to be due to the deterioration in their habitat caused by several factors. Shading by overgrowing shrubs and the closure of the tree canopy, nearby intensive agriculture, urbanisation and use of the forest for recreation are the more important factors threatening and ultimately leading to the extinction of colonies. At poor quality sites there are no or very few nests of Serviformica ants, which initially serve as host ants and are necessary for recolonization by red wood ants. Thus, it is unlikely that these sites will be recolonized. To conserve red wood ants and maintain populations there is an urgent need to undertake certain measures, especially in areas outside nature reserves. For this there is a need to create clearings in the woodland, reduce shading due to overgrowing shrubs and closure of the tree canopy, and mitigate the effects of intensive agriculture, urbanisation and recreation. It is important that the effects of these measures should be monitored.
